What is the meaning of "act" here?

Not too sure but I think it might mean a "rehearsed" performance, as in, one not done under improvisation.

It is just the dictionary meaning form Paper Oxford English Dictionary. Without a context, I do not really know what set refers to.

I think it means a prescheduled performance. An example of how you might use it in this meaning in a sentence is telling a group who is about to perform on stage, "Your act is on in 5 minutes."
