I just wanted to know exactly the meaning of phrase "I sure hope not" or "I sure hope so".

It means that you wish something will happen.

Your question is sort of grammatically correct but it sounds awkward.
A better version would be: I was wondering, what is the meaning of the phrase: "I sure hope not" or "I sure hope so".

Well, it really should be "I was wondering: what is the meaning of the phrase 'I sure hope not'?". Or "I was wondering what the meaning of the phrase 'I sure hope not' is".

"I sure hope not" means "I hope that the preceding is not true".

"I sure hope not." is an emphasis of "I hope not."
"sure" is added to emphasize the sentence "I hope not."
