What's the difference between 'to be supposed to' and 'should'.

"should" is used in advices.
Anyway, I don't know!

Both can mean "to be expected to".

He is supposed to/should arrive at six.
You are supposed to/should know the answer.
It should/is supposed to rain later.

Should carries more certainty.

Parent talking to a child: You should have these dishes done.

Dark clouds on the horizon: It should rain tommorrow.

Though generally they're the same thing.
